38 April 2015 Tablets & Capsules the mixer first, and the liquid is added as the mixer is moving the powder bed. Low-shear mixing is preferred, and the liquid should be added as a fine spray and over a short period. Over-mixing—applying too much shear or prolonging the mixing time—should be avoided. The most common types of silica used as carri- ers are magnesium aluminum silicate, calcium silicate, precipitated silica, and granulated fumed silica. Tabletting aid In addition to silica's ability to im - prove powder flow as a moisture ab - sorbent and glidant, it can help re - solve a number of tabletting problems. It can reduce picking and sticking to tablet punches, improve the me cha - nical properties of tablets (greater hard ness and less friability), increase bulk and tap density of powders (higher fill weights), and to re duce tablet capping, chipping, and lamina- flowability. Blending must generate sufficient shear force to break up the silica agglomerates but be gentle enough not to break up the host parti- cles. Many types of silica are used as glidants and anti-static agents, includ- ing fumed silica, precipitated silica, magnesium aluminum silicate, magne- sium trisilicate and magnesium silicate. Carrier Incorporating liquid ingredients into oral solid dosage forms can be a challenge, but silica enables you to convert liquids into free-flowing powders for this purpose. Examples include vitamin E and liquid flavors for use in tablets. The most useful class of liquid carriers is known as mesoporous silica. These materials have not only a large surface area, but are also highly porous. Optimizing adsorption into the carrier requires proper mixing condi- tions. Generally, the silica is added to tion. Fumed silica and precipitated sil- ica are the most commonly types used as tabletting aids, and the typical use level ranges from 0.1 to 2.0 percent. Film-coating ingredient Film coatings have a variety of roles in oral solid dosage forms, in cluding taste masking, controlling drug release, improving product stability, easing swallowing, and promoting brand recognition. Film coatings are com- monly applied to tablets, pellets, API crystals, granules, and capsules. Silica is used in coatings to stabilize spray suspensions (minimize settling in spray lines and prevent nozzle clogs), improve texture, shorten drying time, increase tablet-edge integrity, and serve as an anti-tacking agent (replac- ing talc) to reduce twinning. Silica use levels for film coatings range from 0.5 to 2.0 percent and the types most commonly used are precipitated silica and fumed silica.
