54 September 2017 Tablets & Capsules sensitive to than others. Steviol gly- cosides also have a lingering sweet- ness, a licorice aroma, and sometimes a delayed onset of sweetness. The intensity of these sensory qualities is determined by the constituents of the steviol glycoside mixture and their concentrations. The more glucose moieties on the steviol, for example, the sweeter and less bitter the glyco- side. Rebaudioside A is the most widely sold steviol glycoside, and its four glucose moieties provide desir- able sensory qualities. In addition, it is typically the second-most abundant glycoside in the Stevia rebaudiana leaf and thus the process to concentrate it is very cost effective. Development Our initial experiments showed that one species delivered a stevia leaf extract that seemed devoid of after- taste and had a very clean sweetness. But because of stevia's inherent antimicrobial nature, myceliation of the stevia leaves took 6 months. That was far too long, and we sought a faster process. The literature about stevia discusses a couple processing methods: glycosy- lation and emulsification. These processes—typically used to create bit- ter blockers—can also be used to miti- gate and alter certain aspects of stevia's taste. At first, my scientist colleagues and I thought the mechanism of action of the solid-state fermentation was an enzymatic glycosylation. Furthermore, glycosylation serves to attach sugar moieties to those already on the mole- cule, which subsequently makes the glycoside taste better. These tech- niques are used on more products than just stevia, and glycosylation can be effected in a handful of ways. To study the issue further, we decided to incubate a steviol glyco- side mixture with the extracellular liquid of a liquid tissue culture. We thought this might effect a glycosy- lation and provide similar sensory qualities to the initial ClearTaste for- mulation. This was indeed the case.
