6 May 2019 Tablets & Capsules research Study finds most oral medications contain adverse-reaction- associated inactive ingredients CAMBRIDGE, MA—According to a study from MIT and Brigham and Women's Hospital, nearly all approved oral forms of medication contain inactive ingredients that may cause allergic reactions or irritations in certain patients. The study, pub- lished in the March edition of Science Translational Medicine, found that the number of publications per year in PubMed containing the search terms "excipient allergy" or "excipient irrita- tion" had increased over the past 3 decades and cited several studies that described patients who'd had allergic reactions to inactive ingredients such as lactose and chemical dyes. The researchers also gathered infor mat ion f r om the Nat ional Library of Medicine's Pillbox data- base to determine the composition of nearly all prescription and over-the- counter medications available in the US. They found that, for most medi- cations, inactive ingredients account for more than half of the mass of each tablet or capsule, and for some, inac- tive ingredients account for 99 per- cent of the product 's mass. T he researchers also found that about 93 percent of medications contain pea- nut oil, lactose, dyes, or other aller- gens and that most contain com- pounds some patients cannot tolerate, such as certain sugars or gluten. W hile the a mo u nt s of the s e ingredients in any one medication are generally too small to trigger s y mptoms, the st udy states that amounts could become substantial for patients simultaneously taking multiple medications. The team is conducting a follow-up st udy in which they are polling healthcare providers to determine how wide- spread adverse reactions may be.
